Abstract:

PURPOSE: To obtain the subject reagent for clinical inspection, etc., capable of determining bilirubin in a specimen by bringing a bilirubin-containing sample into contact with a mixture of an oxidase such as bilirubin oxidase with urea and optically measuring the change in amount of bilirubin.
CONSTITUTION: In this reagent for optically measuring the change in bilirubin amount by reacting bilirubin with an oxidase, a composition containing 100mM potassium hydrogen phthalate (pH5.3), 40mM p-toluenesulfonic acid, 2.0mM N-acetylcysteine, 2.0mM sodium fluoride, 0.002mM human serum albumin and 0.05% polyethylence glycol-p-isooctylphenyl ether is prepeared and 5U/ml of an oxidase such as bilirubin oxidese and 0.5M urea added to the composition to provide the objective reagent for measuring bilirubin, capable of accurately measuring bilirubin in plasma specimen, enabling measurement of bilirubin concentration in a short time after collecting the sample and effective in diagnosis of hemolytic anemia, hemolytio jaundice, obstructive jaundice, etc.
