PURPOSE: To obtain a recombinant strain deleted in a selective marker gene, by transforming a bacterium, etc., with a vector containing a fixed DNA fragment and a specific selective marker gene.
CONSTITUTION: A bacterium, a filamentous fungus, a yeast, etc., are transformed with a vector containing a fixed DNA fragment and a DNA repeat adjoining and dominant two-direction selection marker gene. An acetamidase gene, especially derived from Aspergillus nidulans is preferable as the selection marker gene. The selection marker gene is readily deleted from a recombinant host microorganism by dominant selection Acet.
