Abstract:
PURPOSE: To obtain the subject new gene coding a polypeptide having a nitrile hydratase gene promotor activating ability, capable of mass-producing nitrile hydratase and efficiently producing heterogeneous proteins by transducing heterogeneous genes.
CONSTITUTION: This new regulator gene DNA has an amino acid sequence containing an amino acid sequence expressed by the formula and codes a polypeptide having activating ability of a nitrile hydratase gene promotor. The mass production of the nitrile hydratase is made possible by together putting the regulator gene in a bacterial cell of the same genus Rhodococcus having the same nitrile hydratase containing the promotor region, and high expression of heterogeneous proteins is made possible by transducing heterogeneous genes under the presence of the promotor. The regulator gene is obtained by separating chromosomal DNA from Rhodococcus rhodochrous JI strains, preparing DNA library and screening the DNA library using a probe.
