Abstract:

PROBLEM TO BE SOLVED: To screen a G protein conjugating receptor ligand by contacting a substance with a cell specimen derived from PC12 and having desired G protein conjugating receptor gene and reporter gene and quantitatively determining and comparing the signal of the reporter gene.
SOLUTION: This process determines whether a substance is an agonist of a desired G protein conjugating receptor or not. The objective ligand can be screened by contacting the substance with a specimen composed of a definite number of cells derived from PC12 and containing exogenotes such as a gene coding for the desired G protein conjugating receptor and a reporter gene expressively connected to the promoter region of a gene expressed by the stimulation through G protein, quantitatively determining the detectable signal generated from the reporter gene expressed in each cell of the specimen brought into contact with the substance and the detectable signal generated from the reporter gene expressed in each cell of a specimen free from contact with the substance and comparing the intensity levels of the signals.
