Abstract:
PROBLEM TO BE SOLVED: To provide a high-throughput screening method for identifying a polypeptide with improved biophysical properties (including solubility, high expression and/or stability (high refolding for thermal denaturation, high resistance to a chemical denaturant, high resistance to protease, especially to gastrointestinal protease such as trypsin, and the like)), especially an antibody fragment.SOLUTION: Using high throughput method for screening a polypeptide, a polypeptide (including monomeric human Vand human V) having desired biophysical properties such as solubility, stability, high expression property, monomer property, binding specificity and non-aggregability is identified. The method comprises the steps of: obtaining a phage display library; allowing infection of a bacterial layer with a library phage; and identifying a phage forming a plaque larger than the average on a layer of cell.
