PURPOSE: To obtain a new modified nucleotide for diagnosis, etc., of diseases detecting pathogen, lesion, antigen, etc., containing a reactive group capable of producing a derivative by detectable label, useful for liquid phase nucleic acid assay, etc., of the specific nucleic acid sequence.
CONSTITUTION: This modified nucleotide represented by formula II [R1 is a reactive group capable of producing a derivative by detectable label; R2 is amide bond, thioether bond or disulfide bond; (x) is 1-8] is obtained by a method or the like comprising reacting a pyrimidinenucleotide represented by formula I [R3 is H, methyl, Br, F or I; R4 is H, a blocking group or an acyl capping group having sensitivity to acids and stability to bases; R5 is H or a phosphorus derivative; R6 is H, OH or OR (R is a protective group having sensitivity to acids and stability to bases)] with sulfodichlorides to sulfoamidate the nucleotide, reacting the resultant compound with diamines to introduce amino group and reacting the resultant amino group-containing compound with a protected aminocarboxylic acid.
