Abstract:

PURPOSE: To obtain a new DNA sequence derived from fungi, having a specific base sequence and encoding a phytase capable of converting phytic acid into inositol and phosphoric acid, etc., and providing a phytase good in thermostability, substrate specificity and pH profile.
CONSTITUTION: This new DNA sequence is derived from fungi, comprises a DNA sequence isolated from Talaromyces thermophilus and represented by formula I, a DNA sequence isolated from Aspergillus fumigatus and represented by formula II and a DNA sequence isolated from Aspergillus nidulans and represented by formula III, etc., encodes a polypeptide having the phytase activity or a polypeptide fragment, etc., still having the phytase activity, has improved thermostability, substrate specificity and pH profile and actions of converting phytic acid into inositol phosphate, inositol and inorganic phosphoric acid and is capable of mass-producing a thermostable phytase useful for assorted feeds, etc., according to genetic engineering.
