Abstract:

PURPOSE: To measure α-amylase activity by reaction rate method in high precision, by using a novel amylose derivative having improved affinity for amylase, improved solubility in water, and stability in an aqueous solution, as a substrate.

CONSTITUTION: In measuring α-amylase activity, an amylose derivative having a carboxymethyl group(-CH2COOH) or its salt in each of 6W35 glucose units of amylase obtained by reacting amylose having about 3,000W20,000mol.wt. with sodium hydroxide and monochloroacetic acid in an aqueous solution is used as a substrate, glucoamylase or α-glucosidase is used as a conjugate enzyme, and an amount of glucose formed is measured.
