Abstract:
An adapter suitable for ultra-trace DNA sequencing, and an application thereof. From the 5' end to the 3' end the adapter comprises in turn a Tag sequence, a PolyN sequence, a first hairpin complementary sequence, a first hairpin structure sequence, dUTP, a second hairpin structure sequence, and a second hairpin complementary sequence; the second hairpin complementary sequence is a reverse complementary sequence of the first hairpin complementary sequence; and the 5' end of the adapter has a phosphorylation tag. Hairpin structures are designed in the present adapter such that, when in use, the adapter self-forms the hairpin structures and is ligated to DNA to be sequenced, effectively preventing the problem of low ligation efficiency caused by self-ligation of the adapter when being ligated to DNA to be sequenced. The present adapter is particularly suitable for use in ultra-trace DNA library construction and sequencing, and for laying the foundation for accurate detection of ctDNA.
