Abstract:
Provided are a bacterium for engineering acarbose, a preparation method therefor, and an application thereof. The bacterium for engineering acarbose is an acarbose producing Actinoplanes bacterium or a derivative strain thereof, in which one or both of the following genes are inactivated: (i) a gene M that codes a polypeptide having the sequence shown in SEQ ID NO: 2 or having a sequence identity of at least 80%, 90%, 95%, or 99% with the sequence shown in SEQ ID NO:2; and (ii) a gene N that codes a polypeptide having the sequence shown in SEQ ID NO: 3 or having a sequence identity of at least 80%, 90%, 95%, or 99% with the sequence shown in SEQ ID NO:3. Production of acarbose using the bacterium for engineering acarbose can reduce impurities of a component A and/or a component B in a product.
