Abstract:
The object aims to reveal the cleavage pattern of double-stranded RNA with Dicer, and to provide siRNA which can be cleaved with Dicer so as to produce a specific sequence and a method for producing the siRNA.  Disclosed is hairpin RNA (shRNA) which is characterized by comprising a first target sequence, a first stem sequence, a sequence capable of forming a loop, a second stem sequence, a second target sequence, and an overhung sequence in this order in the 5’→3’ direction, wherein the first target sequence and the second target sequence are annealed to each other in the molecule to form a complementarily bound strand, the first stem sequence and the second stem sequence are annealed to each other in the molecule to form a complementarily bound strand, and one of the first stem sequence and the second stem sequence contains one bulge base that does not anneal to nucleotides in the other of the first stem sequence and the second stem sequence, or DNA encoding the hairpin RNA.
