Abstract:
The present invention provides: a protein containing an altered-type TALE section which exerts excellent binding activity with respect to a given target DNA base sequence with which sufficient DNA binding activity could not have been conventionally achieved; a nucleic acid coding for the aforementioned protein; an expression vector containing said nucleic acid; uses for the aforementioned protein, nucleic acid, and expression vector; and a method for screening the aforementioned protein. Specifically, the present invention provides an isolated altered-type protein exerting binding activity with respect to a target DNA, the altered-type protein containing a Transcription Activator-Like (TAL) Effector section derived from a Xanthomonas bacterium and having a repeating structure of multiple modules, wherein each of the modules has a target base recognition region for recognizing one base constituting the base sequence of the target DNA, and at least one of the modules has an amino acid sequence which was altered so that at least one amino acid residue, which is selected from a group comprising the 2nd to 6th and the 30th to 34th amino acids in an amino acid sequence represented by sequence number 1, has an artificial mutation.
