Abstract:
Provided is a method for capturing a RNA in situ advanced structure and interaction. The method comprises: fixing a protein-mediated RNA interaction in a cell or tissue; perforating a membrane while maintaining the integrity of the cell; degrading a free RNA; performing pCp-biotin labeling at the tail of RNA3' and connecting proximally in situ; performing cell lysis, and then purifying the chimeric RNA containing C-biotin; constructing a strand-specific library; and performing high-throughput sequencing.
