Abstract:
The present invention relates to a method for detecting alkaloid in Coptidis rhizoma. The detection method comprises the following steps: infiltrating a Coptidis rhizoma sample to be detected with an embedding agent and freezing same to prepare a slice; and attaching the slice to a slide, and detecting the alkaloid in the slice by means of time-of-flight secondary ion mass spectrometry. The Coptidis rhizoma sample is fresh Coptidis rhizome product or a dry Coptidis rhizome product which has been subjected to wetting treatment, wherein the solvent used for wetting treatment contains water; and the embedding agent contains a water-soluble mixture of polyethylene glycol and polyvinyl alcohol. According to the method, the time-of-flight secondary ion mass spectrometry is introduced into the detection of the alkaloid in Coptidis rhizoma for the first time without firstly enriching/extracting the alkaloid components in the Coptidis rhizoma sample, so that the operation is simple, the interference is low, the loss of the alkaloid can be effectively avoided, the precision, accuracy, reproducibility and stability of the detection are good, the detection result can be visually presented, and the detection efficiency is high.
