A simple and efficient method of determining a substrate contained in a sample, the method being reduced in interference with the hemoglobin contained in the sample and being applicable to various automatic analytical apparatuses; and a reagent for the determination. The method, which is for determining a substrate contained in a hemoglobin-containing sample, comprises causing an oxidase corresponding to the substrate to act thereon and optically measuring the amount of the resultant hydrogen peroxide with the aid of a peroxidase and an oxidizable color-assuming reagent and is characterized by treating the hemoglobin-containing sample with an anionic surfactant selected among polyoxyethylene alkyl ether sulfuric acid salts, polyoxyethylene alkyl phenyl ether sulfuric acid salts, polyoxyethylene alkyl ether phosphoric acids, polyoxyethylene alkylsulfosuccinic acids, polyoxyethylene alkyl ethercarboxylic acid salts, polyoxyethylene alkyl ethersulfonic acid salts, laurylsulfuric acid triethanolamine, alkylsulfosuccinic acids, and alkyl phenyl ethersulfonic acid salts.
