Abstract:
Disclosed is a method for improving adhesion force of a recombinant mussel foot protein. According to the method, by means of bioinformatics and molecular dynamics simulation, solvent accessibility of protein molecule amino acid residues is calculated, and site-directed mutation is carried out on residue sites. In the present invention, after amino acids at sites N33, N46, and N62 of a wild-type Mytilus coruscus type-3 protein Mcofp-3A, sites G41 and N44 of a wild-type Mytilus galloprovincalis type-3A protein Mgfp-3A, and sites G33, N43, and N76 of a wild-type Mytilus galloprovincalis type-3B protein Mgfp-3B are each mutated into tyrosine that has an adhesion effect on the mussel foot protein, the mutant proteins have no obvious changes in expression, and they have an amino acid sequence set forth in any one of SEQ ID NO.1-8 and have improved adhesion performance with the adhesion force improved by 16%-38% and 12%-82% after liagation for 18 hours in an aqueous solution and a saline solution, respectively, compared with the adhesion force of the original proteins.
