Abstract:

Properties of a protein are modified by the following steps: (a) selecting 1,000 or more pairs of genes orthologous with each other from among genes encoded respectively by genomes of a first microorganism and a second microorganism which is closely related to the first microorganism but different from it in the optimum growth conditions; (b) concerning each gene pair thus selected, detecting amino acid substitutions between the amino acid sequence encoded by the gene of the first microorganism and the amino acid sequence encoded by the gene of the second microorganism and counting the thus detected amino acid substitutions for every amino acid substitution mode to thereby calculate the amino acid substitution frequency; provided that every amino acid substitution count is corrected by subtracting the amino acid substitution count in the opposite direction from it; (c) an amino acid substitution having a frequency at the high-order is identified as an amino acid substitution participating in the optimum growth conditions inherent to the second microorganism; and (d) transferring one or more amino acid substitutions identified in the above (c) into the gene encoding the protein as described above to thereby modify the properties of the protein.
