Abstract:
Disclosed is a method for preparing L-2-aminobutyric acid by means of double enzyme series connection, relating to the field of bioengineering. By respectively culturing recombinant Escherichia coli expressing L-glutamate mutase and L-aspartate-β-decarboxylase, L-glutamate mutase and L-aspartate-β-decarboxylase are obtained. Two enzymes are added to a reaction system at a certain mass ratio, and L-glutamic acid is used as a substrate to carry out an enzyme reaction to prepare L-2-aminobutyric acid. When the amount of L-aspartate-β-decarboxylase is 2 mg/mL, the reaction is carried out for 24 h, and conversion is carried out to generate 8.5 mmol/L of L-2 aminobutyric acid, the molar conversion rate being 85.00%. Compared with chemical production methods, the method has a safe production process and causes no environmental pollution. Compared with a multi-enzyme synthesis system using threonine as a substrate, the substrate is cheaper and the process is simpler.
