Abstract:
The present invention relates to a method for purifying fibrinogen and, more specifically, to a method for purifying fibrinogen, comprising the steps of: (a) (first glycine precipitation) precipitating fibrinogen, which is contained in a solution containing the fibrinogen, by adding glycine thereto until the glycine concentration becomes 1.5-2.5 M, removing a supernatant, and then obtaining a precipitate; (b) (second glycine precipitation) obtaining a lysis solution by lysing, in a lysis buffer, the precipitate obtained by the first glycine precipitation in step (a), precipitating the lysis solution by adding glycine thereto until the glycine concentration becomes 0.2-1.2 M, and recovering a supernatant; (c) (third glycine precipitation) precipitating the supernatant of step (b) by adding glycine thereto until the glycine concentration becomes 1.5-2.5 M, and obtaining a precipitate; and (d) obtaining a lysis solution by lysing, in a lysis buffer, the precipitate of step (c), and performing nanofiltration (NF) thereon by means of a nanofilter. According to the present invention, the novel method for purifying fibrinogen protein is a very safe method since the inflow of pathogenic animal-derived components (virus) is fundamentally intercepted, is very economical and efficient since high-purify fibrinogen protein having a high yield can be purified by a simple process, and can be useful for preventing and treating blood-related diseases, particularly, blood coagulation disorders since there is an advantage of increasing topical effects by having a high purity compared with that of fibrinogen purified by a conventional method.
