Abstract:
A method for qualitatively detecting neutral oligosaccharides in breast milk, successively executing the following steps: (1) centrifuging breast milk, removing upper fat and protein, and taking an intermediate layer of a centrifugal product; (2) adding ethanol and causing same to stand still for a reaction; (3) centrifuging same, and taking a supernatant; (4) loading a sample to a first chromatographic column for adsorption, eluting same with ultrapure water, collecting an eluent, and nitrogen-blowing same for drying; (5) redissolving the dried elution product, loading a sample to a second chromatographic column, eluting same with ultrapure water having 0.1-0.5% of TFA, and collecting an eluent; and (6) assaying the eluent collected in the step (5) by means of liquid chromatography-mass spectrometry, wherein liquid post-column addition is carried out during a liquid chromatography process, and an added reagent is a methanol solution of formic acid. By means of specific selection of an extraction process and of liquid chromatography and mass spectrometry parameters, seventeen types of neutral oligosaccharides in breast milk can be qualitatively detected, and seven types thereof are accurately and qualitatively detected by means of accurate mass spectrum fragment information.
