Abstract:
The presence of colon tumors in a subject is predicted, without performing a bisulfite treatment, by providing in the following order: (a) a step for extracting the DNA in a biological sample collected from a subject; (b) a step for treating the DNA obtained in step (a) using a methylation-sensitive restriction enzyme; (c) a step for amplifying a region encoding the TWIST1 gene, BMP3 gene, NDRG4 gene, or SEPT9 gene, a region from -1 to -1100 from a start codon of any one of these genes, or part of a region from -300 to -500 from a transcription start point of any one of these genes in the DNA treated using the methylation-sensitive restriction enzyme in step (b), and measuring the degree of methylation of one or more CpG sequences within the amplified DNA; and (d) a step for predicting the presence of colon tumors in the subject when the degree of methylation measured in step (c) is at or above a prescribed value, and predicting the absence of colon tumors in the subject when the degree of methylation is less than a prescribed value.
