Abstract:
An observation device 1A comprises a light source 11, a mirror 22, a condensing lens 24, an objective lens 25, a beam splitter 41, an imaging unit 43, and an analysis unit 50. The analysis unit 50 comprises an interference intensity image acquisition unit 51, a first complex amplitude image generation unit 52, a second complex amplitude image generation unit 53, a two-dimensional phase image generation unit 54, a three-dimensional phase image generation unit 55, and a refractive index distribution calculation unit 56. The analysis unit 50 changes the orientation of the reflective surface of the mirror 22 so as to irradiate an observation object S with light from each of a plurality of light irradiation directions, acquires from the imaging unit 43 interference intensity images at a reference position for the respective light irradiation directions, and performs a required process on the basis of the interference intensity images so as to obtain a three-dimensional refractive index distribution of the observation object. This makes it possible to achieve three-dimensional refractive index tomography in which the effect of multiple-scattering light is reduced.
