Abstract:

The present invention relates to the technical field of biology, and relates to an oligosaccharide fluorescent marker, a preparation method therefor, and use thereof. The preparation method of the present invention comprises the following steps: reacting an oligosaccharide with a fluorescent agent in a solvent under the action of a reducing agent to obtain the oligosaccharide fluorescent marker. The oligosaccharide fluorescent marker of the present invention can utilize an amino group in the fluorescent agent to undergo a condensation reaction with a hemiacetal group at a reducing end of the oligosaccharide to form an imine or Schiff base, and then the reducing agent is used to reduce the imine or Schiff base into a relatively stable secondary amine, so that the fluorescent marking at the tail end of the oligosaccharide is realized. The fluorescent group marked at the tail end has little influence on the biological activity of the oligosaccharide, and the marker can have a relatively strong fluorescence intensity under physiological conditions and can stably exist. The oligosaccharide fluorescent marker of the present invention is basically free of toxicity to cells, and the oligosaccharide fluorescent marker can be selectively taken up by oligodendrocyte precursor cells, being suitable for microscopic imaging analysis.
