Abstract:
Provided is optical analysis technology for observing the polarisation characteristics of a light-emitting particle by using a scanning molecular counting method employing optical measurement performed with a confocal microscope or a multi-photon microscope. In the optical analysis technology for observing the polarisation characteristics of a light-emitting particle according to the present invention, excitation light comprising predetermined polarisation components is irradiated in an optical detection region of an optical system while the position of the optical detection region is moved within a sample solution due to the optical path of the optical system of a microscope being altered, and also the intensity of at least one polarisation component of light from the optical detection region is detected. Signals of light-emitting particles are individually detected in the intensity of the at least one polarisation component of the detected light, and polarisation characteristic values of the light-emitting particles are thereby calculated based on the intensity of the at least one polarisation component of the detected signals of the light-emitting particles. It is therefore possible to observe the polarisation characteristics of a light-emitting particle having a low concentration or numerical density in a sample solution.
