Abstract:
It is intended to provide a photofunctional nucleic acid by which the methylation state of cytosine can be clearly determined even in the case of a DNA strand having a consecutive guanine sequence. In a photofunctional nucleic acid of one mode, one base in an oligonucleotide having a sequence complementary to the base sequence of a p53 gene containing the cytosine region, the methylation state of which is to be determined, is substituted by a naphthoquinone compound. This naphthoquinone compound is located at such a position as facing to the cytosine to be determined when the photofunctional nucleic acid is hybridized with the DNA having the base sequence of the p53 gene. The photofunctional nucleic acid is hybridized with the DNA having the base sequence of the p53 gene and then photo irradiated. Thus, the DNA is cleaved exclusively in the case where the cytosine facing to the naphthoquinone compound has been methylated. By analyzing a DNA fragment with the use of polyacrylamide gel electrophoresis, therefor, it can be determined whether the cytosine contained in the DNA has been methylated or not.
