Abstract:
Disclosed in the present invention is a preparation method for an N-acetyl-D-galactosamine trimer precursor. With respect to the preparation of the trimer precursor, a preparation method for compound 4 comprises: adding compound 3, a 4Å molecular sieve and a reaction solvent into a reactor, performing filling and replacing of a protective gas three times, performing stirring, adding enol first and then slowly dropwise adding trimethylsilyl trifluoromethanesulfonate, quenching the reaction by means of an alkali solution when the reaction is finished, and carrying out operations such as extraction and liquid separation, washing, drying and filtration to obtain compound 4; and a preparation method for compound 5 comprises: adding compound 4 and an organic solvent into a reactor, performing stirring, slowly adding an NaIO4 solution into the reaction solution and stirring same for 15 minutes, then adding RuCl2 and stirring same for 1 hour, and performing spin-drying to obtain compound 5. The present invention solves the problems of complex production processes, multiple times of column chromatography purification of products and low yield in the prior art. The present invention achieves further development and updating of production processes for GalNAc.
