Abstract:
Provided are a stem cell-derived gastruloid model, a method for constructing same, and use. The gastruloid model is acquired by means of in vitro construction from a human pluripotent stem cell. The model can simulate biological events of early embryo development and the key structure of embryos to a certain extent, such as the development of the original endoderm, the establishment of the bilaminar embryonic disc, and the appearance of amnion cavity, amnion cells and primitive streak, and has been confirmed in both the protein level and the transcriptome level. The model can well reproduce key features of the embryo from the peri-implantation period to the gastrula stage, and can be produced by batch induction to serve as an early embryo screening model for medicines, environmental toxicants, and the like, thereby providing a safety test for medicine use and the like for certain early pregnant patients.
