Abstract:
Provided is an iPS precursor cell capable of highly efficient transgenesis as a result of stable maintenance cultivation. The precursor cell is prepared by introducing either OCT4, SOX2, or KLF4 to a human somatic cell, and culturing same in a medium including an MEK inhibitor, a GSK3β inhibitor, and a leukemia inhibitor factor (LIF). The MEK inhibitor is, for example, PD0325901. The GSK3β inhibitor is, for example, CHIR99021. The iPS precursor cell is OCT4 negative, NANOG negative, and NESTIN positive.
