組換え抗TNF−αキメラモノクローナル抗体の製造方法及び用途を提供する。ハムスターに最適化したコドンに基づいてCMAB008抗体の軽鎖と重鎖を設計・合成し、真核発現ベクターを構築し、GS遺伝子がノックアウトされたCHO−CR−GS-/-宿主細胞にトランスフェクトし、且つ無血清培養技術で培養し、分離精製し、低免疫原性のCMAB008抗体を得る。
