Abstract:
To provide novel techniques that enable analysis of in vivo protein-RNA interactions more simply and in a shorter time, and further even a small amount of a sample.SOLUTION: Provided is a method for detecting protein-RNA interactions comprising: (1) crosslinking a protein-RNA binding in a test cells; (2) dissolving the test cells after the step (1) to prepare a cell lysis liquid; (3) capturing on beads through target-specific antibodies the target protein-RNA complex in the cell lysis liquid; (4) recovering the beads and treating the captured target protein-RNA complex with endonuclease; (5) treating the target protein-RNA complex on the beads after the step (4) with 5' to 3' exonuclease; (6) treating the target protein-RNA complex on the beads after the step (5) with protease; and (7) recovering RNA from the treatment solution of the step (6) and conducting sequencing analysis.SELECTED DRAWING: Figure 1
