Abstract:
PURPOSE:To enable high manifestation of extraneous gene by replacing a base sequence of part of beta-actin gene promoter of chicken with a gene derived from beta-globin of rabbit. CONSTITUTION:A manifestation vector obtained by removing a gene on 3' end side including splice acceptor sequence in the downstream (on 3' end side) from the midway of intron region in beta-actin gene promoter of chicken and connecting extraneous gene containing splicing acceptor of beta-globin of rabbit thereto and having a hybrid promoter and further having terminator sequence as control sequence for manifestation of extraneous gene is used as a novel manifestation vector. An eucaryotic cell is transformed by a recombinant plasmid having extraneous gene integrated in the downstream of hybrid motor of the vector and cultured.
