Abstract:
PURPOSE:To obtain the title compound, an antitumor antibiotic industrially and advantageously by treating a culture solution of mitomycin C producing microorganism with a reverse-phase adsorbing resin, extracting with ethyl acetate, further passing through a column packed with a reversed-phase adsorbent, followed by concentrating and crystallizing. CONSTITUTION:A culture solution of mitomycin C producing bacterium is treated with a reversed-phase adsorbing resin comprising a polystyrene divinylbenzene-based resin, mitomycin C is adsorbed on the resin, eluted with ethyl acetate, mixed with a phosphoric acid buffer solution having pH6.5-8.0, and distilled. The distillation residue is passed through a column packed with a reversed-phase adsorbent (preferably octadecylated silica gel having 10-200mu particle diameter) under pressure, further eluted with hydrous methanol under pressure, treated with the reversed-phase adsorbing resin, eluted with methanol, further concentrated and crystallized to give the objective mitomycin C.
