Abstract:
The present invention relates to a method for differentiating between live and inactivated bacteria in a whole sample comprising the steps of (i) staining the whole sample with a single membrane-associated dye, (ii) illuminating the stained whole sample with an incident light having an excitation wavelength, (iii) measuring an emission intensity at a range of emitted wavelengths, (iv) creating, for the whole sample, a first emission fluorescence spectra from the plurality of emitted wavelengths, and (v) comparing the first emission fluorescence spectra with an emission fluorescence spectra generated from a sample comprising live bacteria only to determine a spectral shift. Further, the invention also relates to a method for differentiating the relative amount of live and inactivated bacteria in a treated sample comprising staining the whole sample with a single membrane-associated dye; illuminating the whole sample with an incident light at λ excitation; measuring, for the whole sample, (i) the intensity I1 of emitted light at wavelength λ1; and (ii) the intensity I2 of emitted light at wavelength λ2; calculating a ratio I2/I1; and determining the relative amounts of live and inactivated bacteria based on whether the calculated I2/I1 is larger or smaller than a predetermined threshold.
