Abstract:
PURPOSE:To link neocarzinostatin to immunoglobulin while keeping the activity thereof, by linking the neocarzinostatin to the immunoglobulin in the form of neocarzinostatin-mercaptopropionate linked to immunoglobulin-pyridyldithiopropionate. CONSTITUTION:Immunoglobulin is reacted with an N-succinimidyl-3-(2-pyridyldithio)propionate of formula I to give an immunoglobulin-pyridyldithiopropionate of formula II. Neocarzinostatin (hereinafter abbreviated to NCS) is reacted with the compound of formula I to afford a compound of formula III, which is then reacted with a dithiothreitol of formula IV to give a compound of formula V. The compound of formula II is reacted with the compound of formula V to afford the aimed linked substance of immunoglobulin-NCS. The above-mentioned linked substance having selectivity for cancerous cells with retained carcinostatic activity is obtained directly from NCS by easy operation in high yield.
