Abstract:
PURPOSE:To obtain the titled enzyme capable of oxidizing D-amino acid and oxidizing cephalosporin C, by separating a DNA carrying D-amino acid oxidase gene from Trigonopsis variabilis and cloning the DNA to E.coli. CONSTITUTION:The enzyme of formula is produced by extraction and purification of a D-amino acid oxidase capable of oxidizing cephalosporin C from Trigonopsis variabilis CBS 4095(TV); determination of the amino group terminal amino acid sequence; synthesis of a mixture of oligonucleotide having a DNA base sequence of a gene estimated from a part of the determined sequence; labeling of the obtained DNA probe with <32>P; selection and separation of E.coli colony exhibiting complementarity with the DNA probe; separation of plasmid DNA from the separated colony; etc.
