1377318 2-Amino-4(3H)-pteridinones BOEHRINGER MANNHEIM GmbH 25 June 1973 [30 June 1972] 30060/73 Heading C2C Novel compounds I: in which R 1 and R 2 are a C 2-5 straight-chain, branched or cyclic alkyl group (optionally substituted by hydroxy or alkoxy) or an alkenyl, aryl or aralkyl radical and R 1 may also be hydrogen or methyl or NR 1 R 2 form a heterocyclic ring which may contain further nitrogen or oxygen atoms and may be substituted by alkyl, hydroxy, hydroxyalkyl or alkoxy radicals and the salts thereof, are prepared by (1) reacting a compound (II): with glyoxal or with a sodium sulphite addition compound, acetal oxime or hydrazone thereof, (2) condensation of a compound III: in which X is a carboxyl group or amide or ester thereof and Y is amino or halogen with a compound A 1 A 2 A 3 C-NR 1 R 2 in which A 1 is amino, alkoxy or alkylthio, A 2 and A 3 are alkoxy or alkylthio or together represent an imino group or A 1 A 2 A 3 together with the carbon atom represent a nitrile group with the proviso that at least two of the substituents X, Y, A 1 , A 2 and A 3 must contain a nitrogen atom, or (3) reaction of a compound V: in which Z is halogen, amino or optionally alkylated mercapto with an amine H-NR 1 R 2 . Compounds I are diuretics, influencing only the excretion of sodium ions, and form with a carrier or diluent a pharmaceutical composition which may be administered orally or parenterally.
