Abstract:

PURPOSE: To prepare L-phenylalanine in high yield, by culturing a bacterial strain belonging to Bacillus genus and containing a vector in which a gene region containing at least pheA gene is integrated.

CONSTITUTION: A chromosome DNA obtained from a mutant belonging to Bacillus genus and resistant to phenylalanine antagonist is incised together with a vactor DNA using a restriction endonuclease, and the chromosome DNA farction is linked with the incised vector DNA using a ligase to obtain a vector integrated with a gene region contining at least the pheA gene. The vector is integrated into a bacterial strain belonging to Bacillus genus. The obtained L- phenylalanine-producing bacilli are cultured in a conventional medium under aerobic consitions and the accumulated L-phenylalanine is separated from the medium.
