Abstract:
PURPOSE: To prepare new compounds having a remarkable intestinal calcium transport activity.
CONSTITUTION: This is a new compound represented by formula I (each of R1 and R2 is H or methyl; when R1 is H, R2 is not H and when R1 is methyl R2 is not methyl). For example, the compound represented by formula I is prepared by converting a compound represented by formula II as a starting material to a compound represented by formula III, deprotonating the compound with a diethylamidolithium, condensing with a compound represented by formula IV, acetylating, and eliminating reductively by a sodium amalgam to obtain a compound represented by formula V, and removing the protecting groups of 25- and 3β-hydroxy of the compound represented by formula V to obtain a compound represented by formula VI (each of R3, R4 and R5 is H or a hydroxy protecting group), isomerizing optically and/or thermally and removing 1α-hydroxy protecting group.
