Abstract:
PURPOSE: To obtain a new 4-trans-(N-isopropylideneamino)cyclohexanol useful as an important intermediate to produce ambroxol known as an expectorant in bronchi and airway.
CONSTITUTION: The 4-trans-(N-isopropylideneamino)cyclohexanol is expressed by formula I. The compd. of formula I is obtd. by hydrolyzing 4-cis/trans- acetoamide cyclohexanol expressed by formula II, allowing the obtd. 4-cis/trans- aminocyclohexanol expressed by formula III to react with acetone. By this method, 4-trans-(N-isopropylideneamino)cyclohexanol which does not completely include cis isomers can be obtd. with high yield. The ambroxol is N-(2-amino-3,5- dibromobenzyl)-trans-4-aminocyclohexanol.
