Abstract:

PURPOSE: To simplify the acclimatization process of a cultured seedling by supplying carbon dioxide gas and irradiating with light.

CONSTITUTION: The culture liquid used in the present process contains a carbon source such as sucrose at the start of culture. In the regeneration of a shoot, the carbon source is substantially completely consumed. There is essentially no heterotrophic culture after the regeneration step of the shoot. An autotrophic culture of the plant is carried out by the supply of carbon dioxide gas and the irradiation with light. The humidity is lowered by the supply of the carbon dioxide gas. Accordingly, the acclimatization step can be extremely simplified. Since the humidity is lowered from the regeneration step of shoot, a healthy seedling can be prepared.
