To rapidly and simply determine the activity of the subject enzyme that is useful in the early diagnosis of plant soil-borne diseases by using a specific p-nitrphenyllaminary-oligosaccharide as a substrate and allowing the substrate to exist together with glycosidase in the reaction solution.
p-Nitrophenyllaminary-oligosaccharide of the formula ((n) is an integer of 1-4), typically p-nitrophenyl-β-D-laminarytetraoside is used as a substrate and glucosidase is allowed to exist together in the reaction solution to rapidly hydrolyze the linkage of p-nitro-phenol and glucose thereby narrowing the difference in the measuring time caused by the origination of β-1,3-glucanase in the specimens. Thus, the activity of β-1,3-glucanase in plants can simply be determined in a shortened time that is useful for the early diagnosis of plant soil-borne diseases, the elucidation of the development mechanism of corn resistance to pathogens and the like.
