Abstract:
PURPOSE: To provide the subject novel compounds useful as anti-tumor drugs.
CONSTITUTION: The compound of formula (wherein X represents a nitrogen- containing base) in a form of (6RS)-diastereoisomer mixture, or single (6R)- or (6S)-diastereoisomers, for example cyclohexylammonium salt of (6RS)-N5)- methyl-5,6,7, 8-tetrahydrofolic acid, is obtained by adding an alkaline (earth) metal salt, preferably a calcium salt or magnesium salt, of (6RS)-N5- methyl-5,6,7,8-tetrahydrofolic acid into an solvent, preferably water, adding, preferably 2 to 2.5 mole of, a nitrogen-containing base, preferably L-arginine, to the mixture, and then an acid such as sulfuric acid to the mixture to convert said alkaline (earth) metal salt into a slightly soluble salt, separating the precipitated slightly soluble salt by filtration and finally removing the solvent by a mean such as a distillation under reduced pressure.
