PURPOSE: To carry out a sensitive quantitative analysis of a specified component by using a peroxidase-marked substance as the specified component of an analytical object, using H2O2, an aromatic primary amine and a specified nitrogen-containing heterocyclic compound as the substrates and measuring the amount of a pigment generated in the enzymatic reaction.
CONSTITUTION: A specified component as an analytical object is adsorbed on a carrier such as a nitrocellulose membrane and a marked substance prepared by marking a substance (e.g. antibody) capable of binding to a specified component through a specific bonding with peroxidase is made to react therewith to bond the specified component absorbed on the carrier thereto. An enzymatic reaction is then carried out using hydrogen peroxide, an aromatic primary amine compound and one compound selected from formula I (R is substitute; (m) is 0 or 1-5; Z is atomic group required for formation of 5-7 membered nitrogen-containing heterocycle; X is H or substituent eliminatable by reaction with oxidized compound of aromatic primary amine compound), formula II and formula III ((n) is 0 or 1-3) as the substrates, and the amount of a pigment generated in the enzymatic reaction due to peroxidase is measured, thus analyzing the specified component.
