Abstract:

PROBLEM TO BE SOLVED: To obtain an artificial ribonuclease capable of cleaving substrate RNAs at specified positions in a certain and selective way, and useful in molecular biology, biological engineering, pharmacology and the like by binding a polyethylene polyamine residue to a midway site of a DNA oligomer, through amide linkage.
SOLUTION: This artificial ribonuclease is obtained by binding a polyethylene polyamine residue to a midway site of a DNA oligomer, through amide linkage and has the structure of formula I (R' is an amide residue of polyethylene polyamine; (n) is an integer of 1-3; B is a base residue of a nucleic acid constituting DNA). During DNA synthesis, a new phosphoroamidite monomer of formula II (R is an ester residue; DMT is an protective group for a hydroxyl group) is reacted to produce an oligonucleotide of formula III (B' is a base residue, whose functional groups are protected, of a nucleic acid constituting DNA) and the ester residue of the resulting oligonucletide is subjected to aminolysis with polyethylene polyamine to obtain the objective ribonuclease.
