NEW MATERIAL:β-N-acetylhexosaminidase having a molecular weight of 56,000±2,000 and having the following physical and chemical properties. Activity, active to natural substrate (e.g. chitin and N-acetylchitooligosaccharide) and synthetic substrate (e.g. p-nitrophenyl N-acetyl-β-D-glucosaminide and p- nitrophenyl N-acetyl-β-D-galactosaminide) to hydrolyze the substrate; optimum pH and optimum temperature, optimum pH is about 5 and optimum temperature is about 70°C when the substrate is p-nitrophenyl N-acetyl-β-D-glucosaminide and optimum pH is about 4 and optimum temperature is about 60°C when the substrate is p-nitrophenyl N-acetyl-β-D-galactosaminide; isoelectric point, about pH9.
USE: Useful for the structure analysis of composite saccharide.
PREPARATION: A bacterial strain such as Nocardia orientalis IFO 12806 is cultured in a medium containing chitin, etc., and the objective enzyme is separated and purified from the filtrate of the obtained culture product.
