PURPOSE: A method of selecting blood samples for transfusion in a short time in which the activity of guanase is determined through an automated process and the blood having lower activity than a standard value is selected.
CONSTITUTION: Geanase activity is determined through an automated process [initial velocity measurement (the xanthine formation reaction is carried out in the presence of a nonionic surfactant; the uric acid formation reaction is carried out in the presence of an electron carrier such as phenazine methosulfate; measurement theory: shown in reaction equation I ), final concentration measurement (the reactions are carried out in the following 4 steps: xanthine formation, 2 steps of hydrogen formation reactions and coloring reaction; measurement thory: shown in equation II)]. Thus, the blood with a guanase activity of 1.8IU/l (calculated using guanine as a substrate) is selected for transfusion. The process according to equation I can increase sensitivity as well as measurement accuracy. The process according to equation II increase the operation speed and preciseness.
