PURPOSE: To obtain the subject enzyme metal complex capable of remarkably enhancing the activity of a natural type enzyme by pretreating a peptidylamide glycolate lyase with a chelating agent such as EDTA and then bringing the pretreated enzyme into contact with cobalt ions (Co2+).
CONSTITUTION: An amidating enzymic composition prepared from an equine blood serum by the affinity chromatography is concentrated with a collodion bag and then subjected to the gel filtration to separate a peptidylamide glycolate lyase. A chelating agent such as ethylenediaminetetraacetic acid(EDTA) is then added to the resultant enzymic solution to carry out the pretreatment. The pretreated solution is further subjected to the gel filtration to remove metallic ions. An aqueous solution such as CoCl2 is then added and brought into contact with cobalt ions (Co2+). The resultant solution is further subjected to the gel filtration and purified to afford the objective peptidylamide glycolate lyase having the remarkably enhanced activity of the natural type enzyme.
