Abstract:

PURPOSE: To provide the subject vector having a DNA region replicative in the cell of bacteria of genus Rhodococcus, a plasmid region replicative in the cell of Escherichia coli and a DNA region containing a drug-resistant gene and useful for the breeding and improvement of industrially utilizable microorganisms.
CONSTITUTION: The objective composite plasmid vector useful for the breeding and improvement of a bacterial strain belonging to genus Rhodococcus or Escherichia and industrially utilizable for the production of valuable substances is produced by integrating (A) a DNA region originated from plasmid pRC001, pRC002, pRC003, pRC004, etc., and replicative and proliferative in the cell of bacteria of genus Rhodococcus, (B) a plasmid DNA region replicative and proliferative in the cell of Escherichia coli and (C) a DNA region containing a drug-resistant gene into a plasmid by restriction enzyme treatment. A transformed microorganism can be prepared by introducing the vector into a cell of bacteria of genus Rhodococcus or Escherichia.
