Abstract:
PROBLEM TO BE SOLVED: To provide compositions and methods for producing glycoproteins with enhanced sialylation using sialidases, which are enzymes to remove the terminal sialic acid residue from glycoproteins and glycolipids.SOLUTION: A cell line comprises disrupted expression of at least one chromosomally integrated nucleic acid sequence encoding a non-cytoplasmic sialidase. The amino acid sequence of the sialidase is selected from the group consisting of (a) amino acid sequences at least about 96% identical to an amino acid sequence of a specific sequence and (b) amino acid sequences at least about 81% identical to an amino acid sequence of a specific sequence.
