To promote the separation of a DNA such as a partial sequence DNA fragment by embedding a sample in an entangled polymer matrix effective in delaying the movement due to electrophoresis of a biomolecule such as a DNA and carrying out the electrophoresis thereof in a separation medium.
A mixture of a biopolymer specimen which is a related partial sequence DNA with a biopolymer having a higher molecular weight than that of the biopolyme specimen and a solvent is prepared and embedded in a polymer matrix effective in delaying the movement of the biopolymer which is a DNA sequence determining template passing through the matrix when arranging an electric field across the matrix. The matrix containing the mixture embedded therein is arranged in communication with one end region of a slender electrophoretic medium effective in separating the biopolymer when arranging the electric field across the end region of the medium. The electric field is applied across the matrix and the other end region of the medium to increase the ratio of the biomolecule in proportion to the biopolymer entering the electrophoretic medium. Thereby, the biomolecule is efficiently separated by the electrophoresis.
